Beschrijving Abemaciclib mesylaat is een potente en selectieve remmer van CDK4 en CDK6 met IC50 van respectievelijk 2 nM en 10 nM in celvrije assays. Fase 3.

In vitro

LY2835219 is een oraal beschikbare cycline-afhankelijke kinase (CDK)-remmer die zich richt op de CDK4 (cycline D1) en CDK6 (cycline D3) Cell Cycle-route, met potentiële antineoplastische activiteit. LY2835219 remt specifiek CDK4 en 6, waardoor de fosforylering van het retinoblastoom (Rb)-eiwit in de vroege G1-fase wordt geremd. Remming van Rb-fosforylering voorkomt CDK-gemedieerde G1-S-faseovergang, waardoor de Cell Cycle in de G1-fase wordt gearresteerd, DNA-synthese wordt onderdrukt en de groei van kankercellen wordt geremd. Overexpressie van de serine/threoninekinasen CDK4/6, zoals waargenomen bij bepaalde soorten kanker, veroorzaakt deregulatie van de Cell Cycle.

In vivo

LY2835219 verzadigt BBB-efflux met een ongebonden plasma IC50 van ongeveer 95 nM. Het percentage van de dosis in de hersenen voor LY2835219-MsOH is 0,5–3,9%. In zowel een subcutaan als intracraniaal humaan glioblastoommodel (U87MG) onderdrukte LY2835219-MsOH de tumorgroei op een dosisafhankelijke manier, zowel als enkelvoudig middel als in combinatie met temozolomide.

<p>HT29 cells were treated with DMSO control, 1 uM selumetinib (Sel), 5 uM PD0332991 (991), 0.3 uM LY2835219 (219) or serum-free (SF) medium for 48 h. Whole cell extracts were prepared, fractionated by SDS/PAGE and analysed by Western blotting with the indicated antibodies.</p>

SJSA cells were treated with the alternate CDK4/6 inhibitors Ribociclib (LEE011) and Abemaciclib (LY2835219) for 30 h, alone or in combination with Nutlin at 20 µM for 6 h. Immunoblot analysis was performed as in (a). Like Palbociclib (PD0332991), the combination of Nutlin with alternate CDK4/6 inhibitors diminished p53 target gene expression when compared to Nutlin treatment alone.
