 2.55mg/ml (9.93mM) Taking the 1 mL working solution as an example, add 50 μL of 51 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Beschrijving Xylazine (BAY 1470 hydrochloride) is een α2-Adrenergic receptor agonist, gebruikt als sedativum en spierverslapper.

In vivo

Xylazine remt dosisafhankelijk de norepinefrine afgifte en lipolyse bij beaglehonden. Xylazine neigt er ook toe de epinefrinespiegels dosisafhankelijk te verlagen. Xylazine HCl vermindert dosis- en tijdsafhankelijk de minimale alveolaire concentratie (MAC) bij paarden. Xylazine HCl verhoogt dosis- en tijdsafhankelijk de bloedglucoseconcentratie bij paarden. Xylazine induceert variabele bilaterale caudale analgesie die zich uitstrekt van stuitbeen tot S3, met minimale sedatie, ataxie en cardiovasculaire en respiratoire depressie bij staande merries. Xylazine vermindert significant de hart- en ademhalingsfrequentie, systolische, diastolische en gemiddelde arteriële bloeddruk, PCV, hemoglobineconcentratie, arteriële zuurstofinhoud en zuurstoftransport. Xylazine resulteert in een significant verlaagde hartfrequentie, een verhoogde incidentie van atrioventriculair blok en een verlaagd hartminuutvolume en hartindex bij paarden. Xylazine induceert sedatie en selectieve (S3 tot Co) analgesie gedurende ten minste 2 uur bij koeien. Xylazine vermindert significant de hartfrequentie, ademhalingsfrequentie, frequentie van penscontracties, arteriële bloeddruk, PaO2, PCV en totale vaste stoffen concentratie, en verhoogt significant de PaCO2, base excess en bicarbonaatconcentratie bij koeien. Xylazine onderdrukt ernstig het N3-veld (-75%) en schaft het klimvezelveld (-100%) volledig af bij de niet-geanesthetiseerde, gedecerebreerde rat. Xylazine-ketamine injecties onderdrukken ook ernstig het N3-veld (-75%) en schaffen het klimvezelveld (-90%) bijna volledig af bij de niet-geanesthetiseerde, gedecerebreerde rat.
