Biologische activiteit

Beschrijving Donepezil HCl is een specifieke en potente AChE-remmer voor bAChE en hAChE met respectievelijk een IC50 van 8,12 nM en 11,6 nM. Dit product is niet oplosbaar in PBS-oplossing. Gelieve het niet op te lossen in PBS voor toediening.

In vitro

Donepezil HCl remt de carbachol-gestimuleerde toename van de intracellulaire Ca2+-concentratie in menselijke SHSY5Y neuroblastoomcellen op een concentratie-afhankelijke manier, wat aangeeft dat Donepezil muscarinische antagonistactiviteit heeft. Een recente studie toont aan dat Donepezil menselijke navelstrengveneus endotheelcellen (HUVEC's) kan beschermen tegen H2O2-geïnduceerde celbeschadiging. Dit kan nuttig zijn als een potentiële therapie voor oxidatieve stress bij cardiovasculaire en cerebrovasculaire ziekten.

In vivo

Intraperitoneale toediening van Donepezil bij ratten veroorzaakt een dosisafhankelijke toename van speekselvloed en tremor, wat duidelijke cholinerge gedragssymptomen zijn, met een ED50 van 6 μmol/kg. Donepezil blijkt iets minder potent te zijn met een ED50 van 50 μmol/kg na orale toediening. . Wanneer afzonderlijk in vivo toegediend, verbetert de 5-HT(4)-receptorinductor, RS67333 (0,3 en 1 mg/kg) en Donepezil (1 mg/kg) de herkeningsprestaties vergeleken met met zoutoplossing behandelde muizen, terwijl co-administratie van subactieve doses van RS67333 (0,1 mg/kg) en Donepezil (0,3 mg/kg) het geheugen verbetert. Deze verbetering wordt echter voorkomen als er ook een 5-HT(4)R-antagonist (GR125487, 10 mg/kg) wordt toegediend.
