Biologische activiteit

Beschrijving 6-Diazo-5-oxo-L-norleucine (Diazooxonorleucine, L-6-Diazo-5-oxonorleucine, DON), een antibioticum geïsoleerd uit Streptomyces, is een glutaminase-antagonist met een IC50 van ~1 mM voor cKGA (nier-type glutaminase). 6-Diazo-5-oxo-L-norleucine vertoont pijnstillende, antibacteriële, antivirale en kankerbestrijdende eigenschappen.

In vitro

6-Diazo-5-oxo-L-norleucine lijkt positief te zijn in de in vitro chromosoomaberratietest, waarbij het een dosisafhankelijke, statistisch positieve toename van structurele aberraties vertoont bij 4 en 20 uur blootstelling zonder S9 en ook bij 4 uur blootstelling met S9.
In vivo

6-Diazo-5-oxo-L-norleucine wordt geëvalueerd op zijn in vivo clastogene activiteit door het detecteren van micronuclei in polychromatische erytrocyten (PCE)-cellen in beenmerg verzameld van mannelijke muizen die intraveneus werden gedoseerd met 500, 100, 10, 1 en 0,1 mg/kg na 24 en 48 uur. De in vivo micronucleustest onthult een statistisch positieve respons voor micronucleusvorming bij 500, 100 en 10 mg/kg na 24 en 48 uur.
