Biologische activiteit

Beschrijving Vitamin E (D-alpha-Tocopherol) is een vetoplosbare vitamine met krachtige antioxiderende eigenschappen. Het is een krachtige peroxyl radical scavenger en remt niet-competitief de cyclooxygenase activity in veel weefsels, en remt ook angiogenesis en tumor dormancy door onderdrukking van de gen transcriptie van vasculaire endotheliale groeifactor (VEGF).

deze verbinding is in staat om necrose in TC-1 cellen te induceren en heeft antioxiderende effecten tegen stikstofmonoxide.
In Vivo

Vitamin E induceert TC-1 celnecrose in vivo en vermindert het tumorvolume bij TC-1 tumor-dragende muizen. Deze verbinding vermindert myeloïde afgeleide suppressorcellen (MDSC's) in tumoren van TC-1 tumor-dragende muizen en verhoogt de T-cel accumulatie.

  • Methode

    For in vitro cytotoxicity experiments, 1×105 TC-1 cells per well are added to 24-well plates. Eighteen hours later, tumor cells are treated with Vitamin E (0, 25, 50 μM). After 18 hours, apoptotic (Annexin V+ and 7AAD−) and necrotic (Annexin V+ and 7AAD+) cells are measured using PE Annexin V Apoptosis Detection Kit I.
