 4.000mg/ml (11.55mM) Taking the 1 mL working solution as an example, add 50 μL of 80 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Biologische activiteit

Beschrijving Nifedipine is een dihydropyridine calcium channel-blokker, gebruikt om hypertensie te verlagen en angina te behandelen.
Doelen

In vitro Nifedipine veroorzaakt een significante concentratieafhankelijke toename van eNOS-eiwitexpressie door gekweekte humane coronaire arterie endotheelcellen. Deze verbinding antagoniseert L-type Ca+ kanalen die in het hele cardiovasculaire systeem voorkomen, maar blokkeert ook Kv-kanalen, die leden zijn van dezelfde supergenfamilie. Het verlaagt dosisafhankelijk de waarden van [(3)H]-thymidine-incorporatie en het totale cellulaire eiwitgehalte, evenals de niveaus van gefosforyleerde extracellulaire signaal-gereguleerde proteïnekinase (ERK) 1/2, mitogeen-geactiveerde proteïnekinasekinase (MEK) 1/2, en zelfs de fosforylering van Pyk2, in vasculaire gladde spiercellen (VSMC). Deze chemische stof onderdrukt de niveaus van proliferatief celkernantigeen (PCNA) dosisafhankelijk in zowel VSMC als bij ballonletsel van thoracale aorta's in VSMC.
In Vivo Nifedipine (3 mg/kg) verlaagt licht de systolische en/of diastolische bloeddruk of verhoogt de hartslag bij ratten. Deze verbinding (1 μm) produceert een maximale remming van de store-operated pathway in gladde spieren van de choroïdale arteriolen. Deze chemische stof (20 en 40 mg/kg) voorkomt significant HCl-plus-ethanol-geïnduceerde maagslijmvliesbeschadiging en de toename van het gehalte aan thiobarbituurzuur-reactieve stoffen in het beschadigde slijmvlies bij ratten. Het (20 en 40 mg/kg) bevordert dosisafhankelijk de genezing van zweren en remt de toename van het gehalte aan thiobarbituurzuur-reactieve stoffen in het maagslijmvlies met zweren bij ratten.
