Biologische activiteit

Beschrijving VO-Ohpic is een krachtige remmer van PTEN (fosfatase en tensine homoloog) met een IC50 van 35 nM.

In vitro VO-OHpic is een krachtige verbinding met een kleine molecule die specifiek de cellulaire enzymatische activiteit van PTEN remt, wat op zijn beurt stroomafwaartse doelwitten zoals Akt en FoxO3a activeert. De glucoseopname in adipocyten wordt drastisch verbeterd bij PTEN-remming met VO-OHpic. PTEN-remmer versnelt wondgenezing in fibroblasten. VO-OHpic remt de cellevensvatbaarheid, celproliferatie en kolonieformatie, en induceert senescentie-geassocieerde h-galactosidase-activiteit in Hep3B-cellen (lage PTEN-expressie) en in mindere mate in PLC/PRF/5-cellen (hoge PTEN-expressie), maar niet in PTEN-negatieve SNU475-cellen.
In Vivo VO-Ohpic remt significant de tumorgroei bij naakte muizen met xenografts van Hep3B-cellen. VO-Ohpic toegediend aan C57BL6-muizen 30 minuten voorafgaand aan KCl-geïnduceerde asystolische hartstilstand, verhoogt significant de overleving, LVPmax en dP/dt max met aanhoudend voordeel. VO-OHpic verhoogt ook significant de lactaatklaring en verlaagt de plasmaglucosespiegel.

    Cell proliferation is determined by estimating the amount of bromodeoxyuridine (BrdU) incorporation into DNA by a colorimetric immunoassay. 3×103 cells are cultured in 96-well plates with varying concentrations of VO-OHpic for 72 hours. BrdU is added 24 hours before the end of the treatments. Results are expressed as the percentage inhibition of BrdU incorporation over the control.
