SF1670 is een zeer krachtige en specifieke PTEN-remmer met een IC50 van 2 μM.

SF1670 vertoont potente cytotoxiciteit in HBEC, PC-3, H1299 cellen met een IC50 van respectievelijk 5 μM, 10 μM, 44 μM. In een muis aorta ring matrigel angiogenese model stimuleert deze verbinding de angiogene processen. Het vergroot chemoattractant-uitgelokte PtdIns(3,4,5)P3 signalering in neutrofielen en verbetert de neutrofiel functies.

In Vivo

Voorbehandeling met SF1670 (500 nM i.v.) verhoogt de bacteriedodende capaciteit bij neutropene muizen bij zowel peritonitis als bacteriële longontsteking, en vermindert de mortaliteit van neutropenie-gerelateerde longontsteking.

Protocol (uit referentie)

Kinase-assay:^{^[1]}	PTEN Remmingsassay Om de dosis-respons van potenti le PTEN-remmers te bepalen, worden doses van testverbindingen vari rend van 1 nM tot 250 uM (uiteindelijke reactiemixconcentraties) ge valueerd in de algemene PTEN-remmingstest. Om uitgevoerde IC50-gegevens te verkrijgen, worden twee afzonderlijke rondes van de dosis-responstest uitgevoerd. In de eerste ronde wordt de PTEN-activiteit getest in aanwezigheid van een remmer in 10-voudige seri le verdunningen, vari rend van 1 nM tot 250 uM. Zodra het concentratiebereik is bepaald waarin de PTEN-activiteit drastisch verandert, worden twee extra concentratiegegevenspunten binnen dit bereik toegevoegd en wordt de PTEN-remmingstest vervolgens opnieuw uitgevoerd voor de tweede ronde. De PTEN-remming IC50 wordt gepresenteerd als de remmerconcentratie waarbij 50% van de PTEN-activiteit optreedt. Wanneer de test meerdere keren werd uitgevoerd en enigszins verschillende IC50's opleverde, worden deze gerapporteerd als een reeks gevonden IC50's.
Celassay:^{^[1]}	Cellijnen Human brain endothelial cells (HBEC), human prostate cancer cells (PC-3) and human non-small cell lung cancer cells (H1299) Concentraties ~1 mM Incubatietijd 2 hours Methode Cells are plated into 96-well plates in RPMI 1640 medium supplemented with 10% FBS and incubated overnight at 37°C. in an incubator containing an atmosphere of 5% CO₂. The following day, the medium is replaced and cells are starved by placing in 100 uL of serum-free medium for 3 hours. Serially diluted test compounds are added to the wells and incubated with the cells for 2 hours at 37°C. Compounds are tested in a range from 1 mM to 0.1 nM depending on solubility. MTT is added to the wells at a final concentration of 5 μg/ml and incubated with the cells for 3 more hours. At the end of the incubation, the medium is aspirated and the MTT stain in the cells is dissolved by the addition of 100 μL DMSO. Optical density of each well is then measured at 570 nm using a SpectroMax Plus spectrophotometric plate reader. The IC50 determination from the data was made using Prism software.
