Biologische activiteit

Beschrijving ISO-1 (MIF Antagonist) is een remmer van de MIF (Macrophage migration inhibitory factor) D-dopachroom tautomerase activiteit met een IC50 van ~7 μM.

In vitro

ISO-1 remt de cytokine-activiteit in vitro significant. Het remt de afgifte van tumornecrosefactor uit macrofagen geïsoleerd uit met LPS behandelde wildtype muizen, maar heeft geen effect op de afgifte van cytokines uit MIF-deficiënte macrofagen.
In Vivo

ISO-1 verhoogt de overlevingskans aanzienlijk bij ernstige sepsis veroorzaakt door endotoxine of blindedarmligatie en -punctie (CLP). Het kan luchtwegovergevoeligheid (AHR) voorkomen in muis-ovalbumine (OVA)-uitdagingsmodellen. Deze verbinding verminderde ozon-geïnduceerde celrekrutering, cytokine-afgifte en AHR in de muis, maar beïnvloedde de metingen van emfyseem niet. Het beschermt fotoreceptoren en vermindert gliose bij experimentele netvliesloslating.

    RAW 267.4 macrophages were treated with various concentrations of ISO-1 (1-100 μM) 30 min prior to LPS addition. Macrophages were collected 2 h after activation with LPS and washed one time in PBS (pH 7.4). Nuclear extract was isolated using NE-PER Nuclear and Cytoplasmic Extraction Reagents.
