Biologische activiteit

Beschrijving Homoharringtonine (HHT), een plantaardig alkaloïde met antitumorale eigenschappen, remt de eiwittranslatie door de initiële elongatiestap van eiwitsynthese te voorkomen via een interactie met de ribosomale A-site. Homoharringtonine remt omkeerbaar IL-6-geïnduceerde STAT3 Tyrosine 705-fosforylering en verminderde de expressie van anti-apoptotische eiwitten.
In vitro

Homoharringtonine (HHT), a plant alkaloid with antitumor properties, induces p62-mediated autophagy in resistant CML K562G cells, thus promoting autophagic degradation of the BCR-ABL protein.
In vivo

In combination with ABT-199, Homoharringtonine (HHT) significantly prolongs overall survival of primary diffuse large B-cell lymphoma (DLBCL) xenograft-bearing mice compared with single-agent approaches.

    Homoharringtonine (HHT) is used to treat K562, K562G, and siRNA-K562G cells seeded in 384-well plates at a density of 8,000 cells/well, at concentrations of 100, 50, 25, 12.5, 6.2, 3.1, 1.56, 0.78 and 0.39 ng/ml for 48 h, with 0.1% DMSO as the vehicle control. According to the manufacturer's instructions, cell proliferation is assayed using an MTS assay, and the absorbance is measured.
