Biologische activiteit

Beschrijving Capsaicin(Vanilloid) is een actief bestanddeel van chilipepers, planten die behoren tot het geslacht Capsicum. Het is een TRPV1-agonist met een EC50 van 0,29 μM in HEK293-cellen. Deze verbinding is een mengsel van Capsaicin en Dihydrocapsaicin.
In vitro

Als een TRPV-1 agonist remt Capsaicin (Vanilloid) dosisafhankelijk de proliferatie van de HCC-cellijnen PLC/PRF/7, HuH7 en HepG2, wat niet wordt beïnvloed door TRPV1 channel blockers. De combinatie ervan met sorafenib verhoogt significant de onderdrukking van celproliferatie, waardoor een synergetisch effect op hoog niveau wordt bereikt en apoptose van HCC-cellen wordt bevorderd door de versterking van p-ERK en de vermindering van p-STAT3-signalering.
In Vivo

De combinatie van Capsaicin (Vanilloid) en sorafenib verbetert de remming van xenograft tumorgroei bij naakte muizen, zonder duidelijke algemene toxiciteit.

  • Methode

    After seeding cells (5×103 per well) in 96-well plates, they are exposed to capsaicin (vanilloid), sorafenib, or their combination for 72 h. Then, the medium is removed and the wells are washed with phosphate-buffered saline (PBS). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay is performed by adding 20 μL of MTT to the cells for 4 h. Light absorbance is measured.
