 0.950mg/ml (2.50mM) Taking the 1 mL working solution as an example, add 50 μL of 19 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Beschrijving Varespladib (LY315920) is een potente en selectieve humane niet-pancreatische secretoire fosfolipase A2 (hnsPLA) remmer met een IC50 van 7 nM en heeft Fase 3 bereikt.

In vitro Varespladib (LY315920) vertoont een significant remmend effect op sPLA2-activiteit in serum van verschillende soorten, waaronder rat, konijn, cavia en mens, met IC50 van respectievelijk 8,1 nM, 5,0 nM, 3,2 nM en 6,2 nM. In BAL-cellen die zijn uitgedaagd met humane sPLA2, vermindert deze verbinding in doses variërend van 0,1 μM–3 μM de vorming van door humane sPLA2 gemedieerde thromboxaan op een concentratieafhankelijke manier met een IC50 van ongeveer 0,8 μM. In de humane conjunctivale epitheelcellijn (HCjE) remt het (10 μM) de door all-trans-retinoïnezuur (RA) geïnduceerde membraangeassocieerde mucine MUC16-expressie significant met 100% na 24 uur en 99% na 48 uur.
In Vivo Ex vivo, LY315920 in doses variërend van 3 mg/kg tot 30 mg/kg i.v. remt de door humane sPLA2 geïnduceerde afgifte van thromboxaan uit cavia BAL-cellen met een ED50 van 16,1 mg/kg. In Transgene Muizen die Humane sPLA2 tot expressie brengen, elimineert zowel orale als i.v. toediening van deze verbinding (0,3 mg/kg–3 mg/kg) de serum sPLA2-activiteit op een dosis- en tijdsafhankelijke manier.
